The U.S. Food and Drug Administration on Friday approved Roche Holdings AG (OTC:RHHBY) Enspryng (satralizumab) for thyroid eye disease.

The regulatory action provides patients with the sole at-home subcutaneous treatment option currently available for this condition.

Clinical Trial Performance

Phase 3 SatraGO trials formed the foundation for this approval. While the initial SatraGO-1 study failed to reach its primary efficacy goal at 24 weeks, the SatraGO-2 trial demonstrated significant clinical benefits.

In the active disease population, Enspryng drove a 52.9% proptosis response rate compared to just 23.4% for the placebo group.

Furthermore, trial data revealed a 60.7% reduction in double vision among treated patients, far exceeding the 25.8% seen in the control cohort.

The treatment maintained a reliable safety profile, mirroring its established safety record in neuromyelitis optica spectrum disorder without generating any new safety signals.

Understanding Thyroid Eye Disease

Thyroid eye disease presents as a severe autoimmune disorder that triggers complex inflammation around the eyes.

The progressive condition threatens vision and causes debilitating physical changes, including redness, eyelid retraction, bulging eyes, and chronic pain.

Affecting roughly 155 per 100,000 individuals, the rare disease frequently accompanies hyperthyroidism, though it can also strike individuals with normal or underactive thyroid function.

In May, Viridian Therapeutics Inc. (NASDAQ:VRDN) announced positive topline results from its REVEAL-2 Phase 3 clinical trial for elegrobart, a treatment for chronic thyroid eye disease (TED).

The REVEAL-2 trial demonstrated that elegrobart achieved proptosis (bulging eyes) responder rates of 50% and 54% for its Q4W and Q8W dosing regimens, respectively, compared with 15% in the placebo group.

RHHBY Price Action: Roche shares were up 0.93% at $57.31 at the time of publication on Friday, according to Benzinga Pro data.
